Anxiolytic Nootropic
Selank
Selank is a synthetic heptapeptide analogue of tuftsin — a naturally occurring immunomodulatory tetrapeptide (Thr-Lys-Pro-Arg). It was developed by the Institute of Molecular Genetics of the Russian Academy of Sciences and has been studied primarily for anxiolytic (anxiety-reducing) and nootropic (cognitive-enhancing) effects in animal and limited human research models.
Research Focus
- Anxiolytic effects
- Memory and learning enhancement
- Stress response modulation
- Immune system interaction
Key Properties
- Heptapeptide (7 amino acids)
- Tuftsin analogue
- Nasal and injectable forms
- No observed sedative side effects in research
Mechanism of Action
Selank appears to modulate the GABAergic system — the same system targeted by benzodiazepines — but without the sedation, dependence, or withdrawal effects seen with traditional anxiolytics. Research also suggests Selank influences enkephalin metabolism (endogenous opioid-like peptides) and promotes BDNF expression, which has implications for memory consolidation and neuroplasticity.
Anxiolytic Without Sedation
Unlike classical anxiolytics, research models suggest Selank reduces anxiety markers without impairing cognitive performance or motor function. This profile has made it a focus of research into non-sedating anxiolytic mechanisms.
ACTH-Derived
Semax
Semax is a synthetic heptapeptide derived from the adrenocorticotropic hormone (ACTH 4-7) fragment. It was developed in Russia in the 1980s and has been studied for neuroprotective, cognitive-enhancing, and stroke-recovery applications. Unlike ACTH itself, Semax does not stimulate cortisol production — the neuroprotective effects appear to be independent of the adrenal axis.
Research Focus
- Neuroprotection
- Cognitive function enhancement
- Stroke and ischaemia recovery
- BDNF and VEGF upregulation
Key Properties
- Derived from ACTH 4-7 fragment
- Does not raise cortisol
- Nasal and injectable forms
- Short half-life (minutes)
Mechanism of Action
Semax has been shown in research to significantly upregulate BDNF (brain-derived neurotrophic factor), a protein critical for the growth, maintenance, and survival of neurons. It also promotes VEGF expression in the brain, which supports vascular integrity and may be relevant to recovery from neurological events. Studies in rat models of ischaemia have shown reduced brain damage and improved recovery with Semax administration.
Key Pathway
The BDNF Pathway
BDNF (Brain-Derived Neurotrophic Factor) is often described as "fertiliser for the brain." It supports the survival of existing neurons, promotes the growth of new neurons (neurogenesis), and plays a critical role in learning and memory by strengthening synaptic connections (synaptic plasticity).
Why BDNF Matters in Peptide Research
Neuroplasticity
BDNF enables the brain to reorganise neural connections in response to learning or injury.
Memory Consolidation
Long-term potentiation (LTP) — the cellular basis of memory — requires BDNF signalling.
Neuroprotection
Higher BDNF levels correlate with protection against neuronal death from stress and ischaemia.
Both Selank and Semax have been shown to influence BDNF expression — making them of research interest not just for anxiety and cognition, but also as potential supports for neurological recovery protocols.

Regulatory Status in South Africa
Neither Selank nor Semax is registered with SAHPRA as a medicine. They are not currently on the WADA Prohibited List for competitive sport. Both are available for purchase as research peptides in South Africa. As with all unregistered research peptides, they may only be acquired and used within a lawful research context.
